| Experimental Notes | Jurkat T cells activated on anti-CD90 coated glass surface for 5 mins, then fixed with 4% PFA at room temperature for 15 mins. Cells were permeabilised with 0.1% Triton X-100 in PBS for 3 min at room temperature, followed by incubation with the primary antibody diluted in 50 mg/mL BSA in 1×PBS for 30 min. The primary was washed off with large volumes of 1×PBS, and the secondary antibody was incubated for 30 min in 50 mg/mL BSA in 1×PBS. Secondary antibody washed with large volumes of PBS. Samples were post-fixed with 4% PFA for 3 min at room temperature. Imaging was performed in a dSTORM buffer consisting of 20% glucose in 10 mM Tris (pH 8) and 50 mM NaCl, 0.8 mg/mL glucose oxidase, 25 mM cysteamine, and 40 µg/mL bovine catalase. |
